Experimental study on nonalcoholic fatty liver disease treated by medical instrument on hepatic diseases of type DSG in rats

Abstract
AIM: To investigate the primary therapeutic efficacy of the medical instrument on hepatic
diseases of type I DSG in treating nonalcoholic fatty liver disease (NAFLD)of rats.
METHODS: Fifty male SD rats were randomly divided into 5 groups.The control groups (n=20,N1 and N2 group) were fed with normal diet; and the model groups (n=30,M1,M2 and T group) were fed with fat-rich diet consisting of 0.88 standard normal diet+0.10 lard oil +0.02 cholesterol.Therapeutic group (T group) was treated from wk 13 and finished at the wk 20..The levels of serum alanine aminotransferase (ALT)、aspartate aminotransferase (AST)、gamma-glu-tamyl-transpeptidase (GGT)、alkaline phospha-tase (ALP)、total protein (TP)、triglycerides (TG)、total cholesterol (TC)、low-density lipoprotein (LDL)、and high density lipoprotein (HDL) were measured. The hepatic histologic changes were observed with HE and Masson staining.
RESULTS: At the end of the experiment,compared with those in the normal control group,the liver index (t=8.65,p<0.01)and serum ALT(t=4.65,p<0.01)、AST(t=4.74,p<0.01)、ALP(t=3.56,p<0.01)、TP(t=6.35,p<0.01)、TC(t=8.41,p<0.01)、LDL(t=8.86,p<0.01) levels were increased significantly in the model control group,and the hepatic steatosis (z=-4.135,p<0.01)、inflammation(z=-3.752,p<0.01) and fibrosis (z=-4.156,p<0.01)were more serious. Compared with those in the model control groups,the serum ALT、GGT、ALP、TP、TG、and TC levels were all down-regulated in the therapeutic group without significances, while AST(t=-2.404,p<0.05) level was reduced significantly.The degrees of the hepatic steatosis and inflammation were decreased without statistical significances,but fibrosis degrees were lightened markedly(z=-3.101,p<0.01).
CONCLUSION: The medical instrument on hepatic diseases of type I DSG has some therapeutic effect in treating NAFLD of rats.

摘要
目的:初步觀察DSG-Ⅰ型生物信息紅外肝病治療儀治療大鼠非酒精性脂肪性肝病的療效。
方法:♂SD大鼠50只隨機分組,正常對照組20只喂飼普通飼料,模型對照組及近紅外光治療組共30只喂飼高脂飼料(0.88標準普通飼料+0.10豬油+0.02膽固醇)。治療組自13wk始進行近紅外線照射治療,于20wk結(jié)束治療并處死各組大鼠,測定血清ALT、AST、GGT、ALP、TP、TG、TC、LDL和HDL水平,同時觀察肝組織學改變。
結(jié)果:20wk試驗結(jié)束時,與正常對照組相比,模型對照組肝指數(shù)(t=8.65,p<0.01)、血清ALT(t=4.65,p<0.01)、AST(t=4.74,p<0.01)、ALP(t=3.56,p<0.01)、TP(t=6.35,p<0.01)、TC(t=8.41,p<0.01)、LDL(t=8.86,p<0.01) 水平顯著升高,肝組織脂肪變性(z=-4.135,p<0.01)、
炎癥(z=-3.752,p<0.01) 及纖維化程度 (z=-4.156,p<0.01)均較對照組程度重。與模型對照組相比,近紅外光治療組肝指數(shù)、血清ALT、GGT、ALP、TP、TG和TC水平均有所下降,但無統(tǒng)計學意義,而AST有明顯降低(t=-2.404,p<0.05);肝組織脂肪變及炎癥程度較模型組輕,但無統(tǒng)計學意義,而肝纖維化程度較模型組減輕(z=-3.101,p<0.01)。
結(jié)論:DSG-Ⅰ型生物信息紅外肝病治療儀對大鼠高脂飲食非酒精性脂肪性肝病有一定的治療作用。
